RESUMO

The Locus coeruleus (LC) modulates various neuronal circuits throughout the brain. Its unique architectural organization encompasses a net of axonal innervation that spans the entire brain, while its somatic core is highly compact. Recent research revealed an unexpected cellular input specificity within the nucleus that can give rise to various network states that either broadcast norepinephrine signals throughout the brain or pointedly modulate specific brain areas. Such adaptive input-output functions likely surpass our existing network models that build upon a given synaptic wiring configuration between neurons. As the distances between noradrenergic neurons in the core of the LC are unusually small, neighboring neurons could theoretically impact each other via volume transmission of NE. We therefore set out to investigate if such interaction could be mediated through noradrenergic alpha2-receptors in a spiking neuron model of the LC. We validated our model of LC neurons through comparison with experimental patch-clamp data and identified key variables that impact alpha2-mediated inhibition of neighboring LC neurons. Our simulation confirmed a reliable autoinhibition of LC neurons after episodes of high neuronal activity that continue even after neuronal activity subsided. Additionally, dendro-somatic synapses inhibited spontaneous spiking in the somatic compartment of connected neurons in our model. We determined the exact position of hundreds of LC neurons in the mouse brain stem via a tissue clearing approach and, based on this, further determined that 25 percent of noradrenergic neurons have a neighboring LC neuron within less than a 25-micrometer radius. By modeling NE diffusion, we estimated that more than 15 percent of the alpha2-adrenergic receptors fraction can bind NE within such a diffusion radius. Our spiking neuron model of LC neurons predicts that repeated or long-lasting episodes of high neuronal activity induce partitioning of the gross LC network and reduce the spike rate in neighboring neurons at distances smaller than 25 µm. As these volume-mediating neighboring effects are challenging to test with the current methodology, our findings can guide future experimental approaches to test this phenomenon and its physiological consequences.

RESUMO

Centrocestus formosanus is a digenean parasite first described from Asia, which parasitizes Melanoides tuberculata (Gastropoda: Thiaridae) at the first intermediate stage, and different fish species as second intermediate host. C. formosanus was previously recorded in Brazilian states, but never before in the southern region of this country. Recording and identification of digenean species through morphological identification is a taxonomic challenge. In light of this, we use an integrative taxonomic approach to report the occurrence of cercariae and metacercariae of C. formosanus in molluscs and fish, respectively, in an urban park located in southern Brazil. Specimens of M. tuberculata and the fishes Poecilia reticulata and Xiphophorus sp. (Poeciliidae) were collected for screening for parasites and molecular analyses using partial fragments of the 28S rDNA gene. The identification of C. formosanus obtained from molluscs and fish specimens permitted us to partially solve the life cycle of this parasite for the first time in the studied environment, demonstrating the necessity of monitoring and controlling molluscs populations. Nevertheless, our results will support future studies aiming to elucidate the life-cycle of C. formosanus in this region, since all sampled hosts' species are invasive in this environment.

RESUMO

A new species of Cambeva endemic to the Rio Ivaí basin, Upper Paraná basin, is described combining morphological and molecular data. This new species is distinguished from all congeners by characters related to the number of pectoral-fin rays, to the colour pattern of the dorsal and lateral surface of the body, to the presence of diffuse blotches in the ventral surface of body, to the presence of a pelvic-fin and pelvic girdle, to the number of odontodes in the inter-opercular and opercular patches, to the number of dorsal and ventral procurrent rays. In addition, the analysis of mitochondrial DNA sequences identified a satisfactory genetic distance between this new species and its congeners. The new species from the Rio Ivaí reinforces its characteristics as an area of endemism for fishes in the Upper Rio Paraná basin.

RESUMO

The genera Hemigrammus and Moenkhausia have been traditionally diagnosed mainly by the former having lateral line completely pored whereas the latter having a lateral line with a few pored scales. Those features have been used to diagnose species of both genera in the upper Paraná River floodplain. Specimens with the diagnostic features of Moenkhausia bonita, collected in the upper Paraná River floodplain, exhibited different developmental levels of the lateral line, making it difficult to distinguish them from specimens of Hemigrammus sp. We analyzed the gene encoding cytochrome C oxidase I (COI) and intron 1 of the nuclear gene S7 to investigate the genetic similarities between the called Hemigrammus marginatus and M. bonita and to confirm their identities. Molecular sequences of other Moenkhausia species were analyzed for genus delimitation tests. The results reveal genetic similarities of M. bonita specimens with different developmental levels of the lateral line, and also distinguish between M. bonita and Hemigrammus sp. Species delimitation tests revealed that specimens from the upper Paraná River floodplain were M. bonita and were distinct from other Moenkhausia species. The developmental level of the lateral line is not a consistent characteristic that distinguishes between Moenkhausia and Hemigrammus species.

RESUMO

The current analysis investigates whether the uplift of the Serra da Esperança and the Ponta Grossa Arch in the Serra Geral resulted in ichthyofaunistic changes in adjacent basins. For this, we describe the phylogeographic structure among populations of Trichomycterus collected in hydrographic basins in southern Brazil by using partial nucleotide sequences of the mitochondrial gene Cytochrome C Oxidase subunit I. Analyses revealed that the nomenclature Trichomycterus davisi fails to contain the whole genetic diversity range found in the collected specimens and indicates at least six genetic lineages in Trichomycterus. Diagnostic morphological characteristics not associated to T. davisi could be identified in some specimens from the Iguaçu Piquiri haplogroup, indicating the occurrence of species Trichomycterus stawiarski. The lack of morphological differences among the other clades clearly suggests a cryptic species case. Molecular analyses revealed at least five new species besides T. davisi in the hydrographic basins and support the interpretation that genetic structure in T. davisi species complex is explained by tectonic events intrinsic to the areas of influence of Serra da Esperança and the Ponta Grossa Arch which occurred around 1.7 My.

RESUMO

Cophylogenetic studies aim at testing specific hypotheses to understand the nature of coevolving associations between sets of organisms, such as host and parasites. Monogeneans and their hosts provide and interesting platform for these studies due to their high host specificity. In this context, the objective of the present study was to establish whether the relationship between Anacanthorus spp. with their hosts from the upper Paraná River and its tributaries can be explained by means of cospeciation processes. Nine fish species and 14 monogenean species, most of them host specific, were studied. Partial DNA sequences of the genes RAG1, 16S and COI of the fish hosts and of the genes ITS2, COI and 5.8S of the parasite species were used for phylogenetic reconstruction. Maximum likelihood phylogenetic trees of the host and parasite species were built and used for analyses of topological congruence with PACo and ParaFit. The program Jane was used to estimate the nature of cospeciation events. The comparison of the two phylogenies revealed high topological congruence between them. Both PACo and ParaFit supported the hypothesis of global cospeciation. Results from Jane pointed to duplications as the most frequent coevolutionary event, followed by cospeciation, whereas duplications followed by host-switching were the least common event in Anacanthorus spp. studied. Host-sharing (spreading) was also identified but only between congeneric host species.

RESUMO

Cichlid fishes are an important group in evolutionary biology due to their fast speciation. This group depends widely of vision for feeding and reproduction. During the evolutionary process it plays a significant role in interspecific and intraspecific recognition and in its ecology. The molecular basis of vision is formed by the interaction of the protein opsin and retinal chromophore. Long-wavelength sensitive opsin (LWS) gene is the most variable among the opsin genes and it has an ecological significance. Current assay identifies interspecific variation of Neotropical cichlids that would modify the spectral properties of the LWS opsin protein and codons selected. Neotropical species present more variable sites for LWS gene than those of the African lakes species. The LWS opsin gene in Crenicichla britskii has a higher amino acid similarity when compared to that in the African species, but the variable regions do not overlap. Neotropical cichlids accumulate larger amounts of variable sites for LWS opsin gene, probably because they are spread over a wider area and submitted to a wider range of selective pressures by inhabiting mainly lotic environments. Furthermore, the codons under selection are different when compared to those of the African cichlids.

RESUMO

Weanling pigs ( = 108, 21 d of age, 5.82 ± 0.16 kg initial BW) were assigned to a 2 × 2 factorial arrangement of treatments to evaluate the effects of dietary levels of CP (high- and low-CP diets) and dried citrus pulp (DCP; 0% and 7.5%) on growth performance, small intestinal morphology, and hindgut fermentation. Pigs were blocked by initial BW and allotted to 1 of 9 pens, each containing 3 pigs. The high-CP diets consisted of feeding 20% and 21% CP levels throughout phase 1 (0 to 14 d) and phase 2 (14 to 28 d), respectively. For the low-CP diets, CP levels were reduced by 4% units as compared with the high-CP diets in both phases. Crystalline AA were supplied to maintain an ideal AA pattern. Pig BW and pen feed disappearance were recorded weekly. On d 7 and 28 postweaning, 1 pig from each pen was euthanized for collection of small intestinal tissues and digesta from cecum and colon. There were no CP × DCP interactions for growth performance and gut morphology. Although the low-CP diet decreased ADG ( = 0.03) and G:F ( = 0.02) from d 21 to 28 postweaning, overall performance was unaffected by the treatments. On d 7 postweaning, pigs fed the low-CP diet tended to have increased ( = 0.09) crypt depth in the duodenum. Low-CP diets tended to increase ( = 0.06) crypt depth and reduce ( = 0.08) villus:crypt ratio in the jejunum on d 7. Dietary treatments did not affect ileal morphology. On d 7 postweaning, low-CP diets tended to reduce ( = 0.09) cecal total VFA, whereas dietary DCP inclusion tended to decrease ( = 0.07) colonic propionate. Including 7.5% DCP to the diet decreased ( < 0.05) colonic isovalerate and ammonia N concentrations on d 7 only for pigs fed the low-CP diet. On d 28 postweaning, DCP inclusion in low-CP diets decreased ( < 0.05) butyrate, isovalerate, and valerate concentrations in the cecum, as well as isovalerate, valerate, and ammonia N concentrations in the colon. Including 7.5% DCP to the diet increased ( < 0.05) acetate:propionate ratio in the hindgut on both d 7 and 28 postweaning only for pigs fed the high-CP diet. Lactate concentration was unaffected by the treatments. These results indicate that feeding low-CP AA-supplemented diets did not compromise overall growth performance, but slightly increased damage in the gut morphology of weanling pigs. Moreover, adding 7.5% DCP to low-CP AA-supplemented diets shifted the fermentation pattern in the hindgut of weanling pigs by decreasing protein fermentation metabolites.

A study was conducted to evaluate the physical, chemical and sensorial quality and fat acids profile of meat from gilts fed diets containing increasing concentration of ractopamine. A total of 468 gilts with initial weight of 84.77±7.20kg were allotted into 36 pens and fed diets containing 0, 5, 10 or 15mg of ractopamine/kg. After an experimental period of 28 days, two animals from each pen were chosen and slaughtered after 15 hours of fasting. A sample of the Longissimus muscle was collected to evaluate the quality of the pork meat. There was no effect (P>0.05) of ractopamine inclusion in pH, water holding capacity, shear force, color and lipid oxidation. However, a quadratic trend was observed (P<0.05) for cooking loss by increasing concentration of ractopamine in diets. For sensorial analysis of the Longissimus muscle, no differences were noted (P>0.05). No effects were observed (P>0.05) for fat acids profile and saturated and unsaturated fat acids ratio. In conclusion, the increase of up to 15mg of ractopamine/kg concentration in the diet does not change the physical and sensorial characteristics and the fat acids profile of meat from gilts slaughtered at 110kg of body weight.

An economic evaluation was done in order to evaluate the effect of the inclusion of 0, 5, 10 and 15 percent of citrus pulp (PC), with or without the addition of the enzyme complex, on the performance of 72 pigs from 25.71±7.51 to 98.81±10.64kg of body weight. The animals were slaughtered when they reached 98.81kg. The purchase value of the animals and feed costs (R$) were discounted from the carcass value, including subsidy rates (R$). A linear increase (P= 0.0190) was observed, while linear reductions were observed for the gross receipts (P=0.0328) and net (P = 0.0013) partials, due to the inclusion of PC in the diet with or without the enzyme complex. Considering the evolution of corn, soybean meal and pig prices during 2008, there were some 12 different scenarios for partial and net income determined and in all of them, linear reductions were observed due to the inclusion of PC in the diet with or without the enzyme complex. The use of PC with or without the addition of the enzyme complex was unable to reduce the costs of feed.

The effects of glutamine, poliunsatured fatty acids and cellular wall of yeast (CWY) under the structure and ultra structure of the small gut and the performance of the piglets were studied. Forty five piglets weaned at 21 days were used to test the following treatments: T1 - basal diet; T2 - basal diet + 1 percent of glutamine; T3 - basal diet + 0,2 percent of CWY; T4 - basal diet + 5 percent of fish oil. At seven and 14 post weaning days, five piglets of each treatment were slaughtered. The height, density of villus and depth of small gut crypts were not altered by the inclusion of additives. The effect of age was observed, showing a reduction in the height and density of villus and depth of crypts after weaning. In duodenum and jejunum higher values were observed in the relation villus:crypt, which increased with the post wean age. There was a decrease in the height of microvillus of the duodenum at 7 days and an increase of the width of the microvillus of jejunum at 14 days after wean. The area of the apical surface of the enterocytes was not altered by the studied factors. The studied additives were not efficient to prevent the atrophy of the intestinal mucosa of the jejunum, since they did not interfere on its ultra structure. Piglet performance was not affected by the additives included in the diet.

RESUMO

An assay with 36 swine initially weighting 83.7±5.1kg body weight (BW) was carried out to evaluate the effects of the use of different dietary citrus pulp levels, 0, 10 percent, 20 percent, and 30 percent, upon digestive organs weights, carcass characteristics, and meat quality of animals subjected to qualitative feed restriction program, and slaughtered at 130kg BW. Linear response (P<0.05) on the weight of stomach, colon, and liver were observed. Negative linear responses (P<0.05) on final BW and carcass yield relative to citrus pulp addition to the diet were observed. Although a negative linear effect (P<0.05) of increasing dietary levels of citrus pulp had been observed on ham weight, yield of this by product was not affected (P>0.05). Higher levels of citrus pulp neither decreased backfat thickness nor increased amount of lean meat, indicative that qualitative feed restriction was not efficient. Positive linear effect (P<0.05) on pH measured 24 hours after slaughter and negative linear effect (P<0.05) on color characteristics as function of citrus pulp dietary levels were verified. Citrus pulp addition in qualitative feed restriction program may not be effective. As no deleterious effects upon meat qualities were observed, citrus pulp can be used as an alternative feedstuff for finishing swine.

A total of 50 barrows were used to evaluate qualitative feed restriction on digestibility of dietary components, weight of organs of digestive tract, and composition and production of feces. Five experimental diets, with increasing levels of qualitative feed restriction of 0, 5, 10, 15, and 20 percent were used. There was linear reduction (P<0.001) of all digestibility coefficients on the levels of qualitative feed restriction, except for acid detergent fiber, which presented a quadratic response (P<0.05). Contents of total solids (P<0.01), volatile solids (P<0.05), and total minerals (P<0.001) in the feces augmented with increasing levels of qualitative feed restriction, whereas the levels of K (P<0.05), Cu (P<0.01), N, P, Na, Ca, Mg, Fe, and Zn (P<0.001) presented a quadratic response. Daily feces excretion; and contents of total solids, volatile solids, total minerals, and levels of N, P, K, Mn, and Cu (P<0.001) and Ca, Na, Mg, and Fe (P<0.05) presented a clear increase in function of the qualitative feed restriction level. Qualitative feed restriction can be an alternative for the disposal of agribusiness waste, providing good crop-fertilizing by the use of swine feces.

The cost of inclusion of citrus pulp was evaluated in a qualitative feed restriction program using performance data of 36 pigs of Topigs lineage (from 83.7±5.1kg to 129.8±1.9kg) fed with crescent levels of citrus pulp (0, 10, 20, and 30 percent). The animals were slaughtered when they reached 130kg. The value paid for animal and its feeding cost were discounted from the carcass value (R$), according to each treatment. No effect (P>0.05) of the inclusion of citrus pulp on ration cost and gross income was observed. A negative linear effect (<0.05) on partial net income with the increasing participation of the citrus pulp in diets was found. The decreasing trend in the partial net income was caused by the reduction on carcass weight of animals fed on crescent levels of citrus pulp. Considering the increase on corn, soybean, and swine prices from June 2005 to May 2006, 12 different sceneries were determined for partial net income analysis. In all situations, linear reductions on partial net income were observed as a function of citrus pulp inclusion, evidencing that the ingredient was not efficient in promoting profits for producers.

RESUMO

The Glasgow Coma Scale (GCS) and the Jouvet Coma Scale (JCS) have been evolved for assessing the depth and duration of impaired consciousness and coma. The analysis and the utilization of these scales have showed that they are complementary. The GCS is more sensitive when there is a more intense loss of consciousness, whereas the JCS shows its sensitivity better in the states close to normal. This study was aimed to compare the results obtained from the evaluation of the consciousness level by the utilization of the two scales. The comparison was done within a prospective study with 48 patients, all of them over 18 years old, interned in three intensive care units of different hospitals in the city of São Paulo. The evaluations were done daily by the researchers and the scales applied in sequence totaling 5 minutes. Each scale was applied in 106 evaluations, and the results showed a statistically meaningful difference between the GCS and the JCS as to the indication of alteration in the consciousness levels. In 37.74% of the evaluations done with the JCS there was an indication of alteration in the consciousness level, whereas with the GCS the alteration was present in only 23.58% of the evaluations. Another important observation about the utilization of both scales was that people whose scores were between 9 and 10 in the GCS had had an stronger indication of alteration of consciousness level by the same scale, while those with scores between 12 and 15 had a stronger indication of alteration in the consciousness level by JCS. When using GCS there has been the application of the non-testable (NT) in 20% of the evaluations. This did not occur when using the JCS. However it is believed that specific conditions of that particular group might have led to that result as well as specific characteristics of groups of patients might favor the utilization of different scales to evaluate the consciousness level. Therefore the final choice between such scales should consider the conditions and the peculiar characteristics of the clientele to be evaluated and not individual or health department services preferences.
